How It Works
Samples are applied to the pre-printed, pre-blocked capture antibody arrays contained in up to eight separate wells on a coated glass slide. The target cytokines from the sample are bound by the capture antibodies during incubation; a second biotin-labeled detection antibody is then added which recognizes a different epitope on the target cytokine. The cytokine-antibody-biotin complex can be visualized using SilverQuant® chromogenic reagents, which allow for the detection of any biotinylated molecule. SilverQuant® chromogenic reagents use a patented, gold-catalyzed silver deposition technology to generate light grey-to-black spots that are visible to the naked eye and can be scanned with the Gentel Proteomics Multi-System™ typically achieving measurably improved sensitivity compared to fluorescence detection. Results are obtained in less than four hours.
Compatible sample types include serum, plasma, cerebrospinal fluid, cell and tissue lysate, and conditioned media. SilverQuant® Profiling Antibody Arrays use the same cytokine-specific antibody pairs as the SilverQuant® Quantitative Antibody Arrays. Twenty two arrays are available in either 4- or 8-sample kit configurations. Cytokines are grouped according to application and antibody cross-reactivity. Corresponding ELISAs, antibody pairs, and protein standards are also available upon request.
