Figure 1. PATH® slides demonstrate higher sensitivity when compared to conventional porous nitrocellulose slides. Titration sandwich immunoassay measurements of purified recombinant IL-1β human cytokine measured between 5 pg/mL to 50,000 pg/mL. The blank results from fluorescence signal in the absence of antigen. σBlank is the standard deviation of the blank. All five dilutions plus blank were performed on a single slide. Twenty replicates per dilution were printed and tested. Experimental parameters for the conventional nitrocellulose slide followed the manufacturer's recommendations. Data where the background signal matched or exceeded actual signal was not plotted.

Figure 2. Four to five logs of dynamic range on PATH® slides. Arraying of cytokine-specific mouse monoclonal antibodies was done with solid pins using an Affymetrix 417 Arrayer. Multiplexed human cytokine sandwich immunoassays on PATH® were done using various concentrations of purified antigen, followed by biotinylated antibodies, then detected using Cy3-labeled Streptavidin. Slides were scanned using a ScanArray™ 4000 laser scanner (Laser Power = 90 PMT/Gain = 75, PerkinElmer Life Sciences).
